摘要 本文利用聚多巴胺(PEG)作为载体来实现人参皂苷Rg1的负载,以期增强其生物活性并提高其稳定性。实验中采用了一系列的方法来优化PEG-Rg1的合成过程,包括不同时间、不同pH值、不同浓度以及不同温度。在优化的过程中,我们发现了一个最佳条件——在pH=7.4、温度=50℃、时间=2h 下使用20mg/ml Rg1浓度进行PEG-Rg1复合物的形成。此外,通过SDS-PAGE和HPLC分析可以证明该复合物在形成过程中保留了大部分原始Rg1的生物学性质。此外,通过对该复合物具有保留原始Rg1生物学功能的实验表明:PEG-Rg1具有微量氧化应力诱导凋亡作用和DNA修复作用。因此,将人参皂苷Rg1装载到PEG上是一个理想的方法来增强其生物学功能并提高其耐久性。 Introduction (引论) Ginsenoside Rg 1 is a kind of active component extracted from Panax ginseng, which has been widely used in traditional Chinese medicine to treat various diseases including cancer and cardiovascular diseases [1]. In recent years, with the development of biotechnology, researchers have found that Rg 1 has some biological activities such as anti-oxidation and DNA repair [2]. However, due to its poor stability, the application of Rg 1 in clinical practice is limited. In order to improve its stability and enhance its biological activity, it is necessary to find an effective way to load it. Polyethylene glycol (PEG) has been widely used as a carrier for drug delivery due to its excellent biocompatibility and water solubility [3]. PEG can form stable complexes with drugs by hydrophobic interactions or hydrogen bonds [4], which can protect drugs from degradation and enhance their bioavailability.
