Research on Non-destructive DNA Extraction Method of Orthoptera Insects
Abstract: Orthoptera insects are important agricultural pests, and their DNA extraction is of great significance for gene analysis, genetic research and molecular biology analysis. This paper introduces a non-destructive DNA extraction method applied to the samples of orthoptera insects. The method uses low concentration acid as processing agent to ensure the integrity of the samples. In addition, PicoGreen DNA quantitation kit is used in DNA concentration determination to accurately measure the content of DNA in the samples. Finally, PCR is used to verify whether the obtained DNA can be used for further molecular biological analysis. By performing non-destructive DNA extraction on different body parts samples and determining the differences between parts, it was found that there were differences between non-destructive DNAs. Therefore, this non-destructive DNA extraction method introduced in this paper will lay an ideal platform for experiments related to genetics, bioinformatics, expression molecular biology and other related fields of orthoptera insects.
