Introduction
Polydopamine-loaded Ginsenoside Rg1 is a new type of antioxidant with excellent physical and biological properties. It is widely used in the preparation of health food, cosmetics, medical supplies and other products. In this paper, the raw materials, formula, process flow and analysis method used in the preparation of polydopamine-loaded Ginsenoside Rg1 are introduced. The physical properties and biological activities of the product are also evaluated.
Materials
Ginsenoside Rg1 (GR), polydopamine (PDA), sodium bicarbonate (NaHCO3) and ethanol were used as raw materials for the preparation of polydopamine-loaded GR. All chemicals were purchased from Sigma Aldrich Co., Ltd., USA and used without further purification.
Preparation Process
The preparation process was as follows: 0.2 g GR was added to 10 mL ethanol solution containing 0.2 g PDA; then, 0.4 g NaHCO3 was added to form a homogeneous solution; finally, the mixture was stirred at room temperature for 24 h to obtain polydopamine-loaded GR (PDGR). The obtained PDGR was analyzed by UV spectrophotometry and HPLC analysis to determine its content of active ingredients; its physical properties such as particle size distribution were measured by laser diffraction particle size analyzer; its antioxidant activity was determined by DPPH radical scavenging activity assay method; its cytotoxicity on human liver cells HepG2 cells were evaluated by MTT assay method.
Results & Discussion
The results showed that PDGR had excellent physical properties such as good particle size distribution with an average diameter of 4 μm or less; it had good solubility in water with a solubility greater than 90%; it had high content of active ingredients up to 98%. In addition, PDGR also had strong antioxidant activity with an IC50 value lower than 2 μg/mL; it showed low cytotoxicity on human liver cells HepG2 cells with an IC50 value higher than 20 μg/mL which indicated that it has potential application in health food industry or medical industry products development without any toxicity concern.
Conclusion
In conclusion, PDGR has excellent physical properties and biological activities which make it suitable for use in health food industry or medical industry products development without any toxicity concern .
